Their ligands, CD155, CD111, CD112, CD113, and PVRL4 are expressed on APCs or tumor cells. TIGIT delivers inhibitory signals through ITIM motifs and ITT-like motifs. The ITT/ITT-like motif of CD226 plays a role in its activating signaling.
